Cocaine addiction is widely considered to be the most severe kind of drug abuse, with a vast number of people dying in this country each year from the consequences of such addiction. Cocaine abuse and dependence also lead to a plethora of non-fatal health problems. Although the mechanisms by which cocaine exerts its pharmacological effects are not fully understood, it is now widely accepted that the major site of action of this drug is in the nucleus accumbens (the ventral striatum). Although there is extensive knowledge about the signaling pathways that mediate the actions of opioids in the nucleus accumbens, the same is not the case for drugs of abuse such as cocaine. The research in this grant proposal is aimed at studying the role of an enzyme called adenylyl cyclase in the nucleus accumbens in response to cocaine and to determine which subtypes of receptors within this area are activated by cocaine. Based on extensive work done in the PI’s laboratory, it is hypothesized that cocaine activates the D1-class of dopamine receptors and that in this way the enzyme adenylyl cyclase is activated. This will lead to an increase in the levels of the second messenger cyclic AMP. The purpose of these studies is to determine the role of cyclic AMP in the actions of cocaine in the nucleus accumbens, using both a neuronal cell culture model and in vivo methods.
